Il protagonista di "Assassin's Creed: Brotherhood" sarà di nuovo Ezio Auditore da Firenze (antenato fiorentino di Desmond Miles), già presente nel capitolo precedente della serie. Ubisoft ha annunciato una innovativa modalità multigiocatore che permetterà di personalizzare i metodi di assassinio sviluppati da Ezio e persino di combattere per i Templari, scegliendo quindi dove schierarsi; il gioco ha come sfondo la città di Roma dove il protagonista dovrà guidare la confraternita di assassini alla ricerca dei capi dell'ordine templare, uscito vittorioso dallo scontro con il Gran Maestro dell'Ordine Templare: Rodrigo Borgia, che ora si vuol vendicare. È stato, inoltre, annunciato che la campagna in giocatore singolo ha una durata di 15 ore.
